Product Description
The DC101-CP089 monoclonal antibody is a recombinant, Fc-engineered chimeric version of the original DC101 antibody. The variable domain sequences are identical but the constant region sequences have been switched from rat IgG1to mouse IgG2a, κ for use in murine models. Additionally, DC101-CP089 contains LALA-PG mutations in the heavy chain Fc fragment rendering it unable to bind endogenous murine Fcγ receptors or C1q to induce antibody-dependent, cell-mediated cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC) while preserving VEGFR-2 blockade. This modification prevents off-target depletion of VEGFR-2-expressing endothelial cells via immune effector functions, focusing activity on signaling inhibition. Species-matched chimeric antibodies demonstrate reduced immunogenicity and formation of anti-drug antibodies (ADAs) compared to xenogenic antibodies in animal models. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies

VEGFR-2 is a member of the tyrosine protein kinase family. Upon binding to its ligand VEGF, VEGFR-2 plays key roles in vascular development and permeability. The DC101 antibody has been shown to inhibit VEGFR-2 signaling in vivo.
