  • Product Description

    The MAR1-5A3-CP056 monoclonal antibody is a recombinant, chimeric version of the original MAR1-5A3 antibody. The variable domain sequences are identical to clone MAR1-5A3, but the constant region has been converted from mouse IgG1 to mouse IgG2a. MAR1-5A3-CP056 also contains Fc silencing mutations rendering it unable to bind to endogenous Fcγ receptors like therapeutic anti-IFNAR-1 antibodies such as Anifrolumab. These mutations prevent Fc-effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.

    MAR1-5A3-CP056 antibody reacts with mouse IFNAR-1 (IFN alpha/beta receptor subunit 1). IFNAR-1 is co-expressed with IFNAR-2 on most cell types and together these two subunits make up the heterodimeric Type I IFN Receptor complex. Type I IFNs (IFN-α/β) bind to the Type I IFN Receptor complex to induce cellular responses including induction of anti-viral, anti-microbial, anti-tumor, and autoimmune responses as well as to regulate activation, proliferation, and differentiation. A growing body of evidence supports the roles of type I IFNs in the immunopathogenesis of autoimmune diseases such as systemic lupus erythematosus (SLE), Sjögren’s syndrome, myositis, scleroderma and rheumatoid arthritis and other interferonopathies. The MAR1-5A3 antibody has been demonstrated to inhibit Type I IFN receptor signaling in vitro and in vivo.
