This non-therapeutic biosimilar antibody uses the same variable regions as the therapeutic antibody Brentuximab, making it ideal for research use. Brentuximab is a humanized IgG1 monoclonal antibody that targets CD30, also known as TNF Receptor Superfamily Member 8 (TNFRSF8), D1S166E, or Ki-1. The CD30 receptor is a 120 kDa glycoprotein antigen that is highly expressed in CD8+ and CD4+ lymphocytes. The CD30 receptor’s ligand is a membrane-bound cytokine, CD30L, that is expressed on activated granulocytes and lymphocytes. The binding of the CD30 receptor to its ligand CD30L activates the receptor activity that leads to the recruitment of TNF-related factors (TRAF1, TRAF2, and TRAF3) and signaling complex formation, thereby stimulating the anti-apoptotic and pro-survival pathways (NFkB and MAPK) in tumors. Besides activated lymphocytes, the CD30 receptor is expressed in eosinophils, B cells, NK cells, dendritic cells (DC), and T regulatory cells (Treg), and its lack in peripheral blood cells makes it an attractive therapeutic target. Brentuximab is the antibody component of the brentuximab vedotin (BV) antibody–drug conjugate (ADC) that is reported to deliver the microtubule-disrupting drug monomethyl auristatin E (MMAE) to CD30+ cancer cells in studies involving Hodgkin lymphoma (HL), systemic anaplastic large cell lymphoma (sALCL), cutaneous T-cell lymphomas, primary cutaneous anaplastic large cell lymphoma (pcALCL), peripheral T-cell lymphoma (PTCL), and mycosis fungoides (MF).
