Product Description
The RM134L-CP233 antibody is a chimeric version of the original RM134L antibody. The variable domain sequences are identical but the constant region sequences have been switched from rat IgG2b to mouse IgG2a. Additionally, the constant region includes the Fc silencing mutation variant LALA-PG rendering it unable to bind endogenous murine Fcγ receptors or C1q to induce antibody-dependent cell-mediated cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). This is useful to study the effects of OX40L neutralization without FcγR/complement‑mediated cell killing, immune activation, or Fc‑driven biodistribution artifacts enabling clean mechanistic dissection. Fc-silent antibodies can also improve in-vitro assays by reducing non-specific signaling and background. The LALA-PG variant has demonstrated significantly reduced effector function, C1q binding and C3 fixation compared to other common silencing mutations while retaining favorable biophysical and manufacturing properties and preserving FcRn-mediated half-life. Species-matched chimeric antibodies result in less immunogenicity and formation of anti-drug antibodies (ADAs) than xenogenic antibodies in animal models and the highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.
The RM134L monoclonal antibody reacts with mouse OX-40L (also known CD252, CD134L, TNFSF4). OX-40L is a 35 kDa member of the TNF superfamily that is expressed on activated B cells, dendritic cells, vascular endothelial cells, mast cells, and natural killer cells. OX40L is the ligand for OX-40 (CD134). OX-40 signaling regulates both CD4 and CD8 T cell clonal expansion, provides a costimulatory signal to an antigen-reacting naive T cells prolonging proliferation, augments the production of several cytokines including RANTES, IL-2, IL-3, and IFN-g and plays an important role in immunotolerance. In vivo treatment with the RM134L antibody has been shown to block the interaction between OX40L and OX40.
