Product Description
The G23-8-CP240 monoclonal antibody is a recombinant, chimeric version of the original G23-8 antibody. The variable domain sequences are identical but the constant region sequences have been switched from Rat IgG1, κ to mouse IgG2a, κ for use in murine models. Additionally, G23-8-CP240 includes the Fc silencing mutation variant LALA-PG, rendering it unable to bind endogenous murine Fcγ receptors or C1q to induce antibody-dependent cell-mediated cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). This results in cytokine neutralization without Fc mediated depletion or immune activation like the Fc-silent therapeutic monoclonal antibody Risankizumab. The LALA-PG variant has demonstrated significantly reduced effector function, C1q binding and C3 fixation while retaining favorable biophysical and manufacturing properties and preserving FcRn-mediated half-life. Species-matched chimeric antibodies demonstrate reduced immunogenicity and formation of anti-drug antibodies (ADAs) compared to xenogenic antibodies in animal models. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.
The G23-8-CP240 monoclonal antibody reacts with the p19 subunit of mouse IL-23. IL-23 is a heterodimeric cytokine composed of two disulfide-linked subunits, a p19 subunit that is unique to IL-23, and a p40 subunit that is shared with IL-12. IL-23 is secreted by activated dendritic cells and macrophages. IL-23 has been shown to enhance IFNγ production by memory T cells. Additionally, mouse IL-23 induces the proliferation of memory T cells (but not naive T cells), whereas IL-12 has no effect on memory cells. Mouse IL-23 (but not IL-12) can also activate mouse memory T cells to produce the potent proinflammatory cytokine IL-17. IL‑23/Th17 signaling drives chronic inflammation in autoimmune conditions such as plaque psoriasis, psoriatic arthritis, Crohn’s disease, IBD and ulcerative colitis. The G23-8 antibody can specifically neutralize mouse IL-23 bioactivity with no effect on IL-12 p70 bioactivity, neutralizing a key upstream driver of the Th17 inflammatory axis.
