Product Description

The IM7-CP238 monoclonal antibody is a recombinant, chimeric version of the original IM7 antibody. The variable domain sequences are identical but the constant region sequences have been switched from rat IgG2b to murine IgG2a for use in murine models. Additionally, the constant region includes the Fc silencing mutation LALA-PG, preventing Fc-dependent cellular depletion, phagocytosis, and complement activation. Fc-silencing enables selective blockade of CD44 function while avoiding Fc-mediated killing or unintended modulation of CD44-expressing immune and healthy cells. Species-matched chimeric antibodies support repeat and long-term dosing studies by reducing anti-drug antibody responses that can accelerate antibody clearance, compromise sustained target neutralization and confound interpretation of in vivo efficacy and safety findings. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.

The IM7 monoclonal antibody reacts with human and mouse CD44 also known as Hermes, HCAM, and Pgp-1. CD44 is an 80-95 kDa glycoprotein that is expressed on all leukocytes, endothelial cells, hepatocytes, and mesenchymal cells. As an adhesion molecule, CD44 participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, and hematopoiesis. CD44 is a receptor for hyaluronic acid and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs) and is involved in tumor metastasis. Targeting CD44 has been shown to reduce the malignant activities of various neoplasms. Interestingly, high levels of the adhesion molecule CD44 on leukemic cells are essential to generate leukemia. IM7-CP238 neutralizes CD44 in vivo and is useful for CD44 blockade in mouse disease models while minimizing Fcγ receptor engagement, complement activation, and depletion of broadly CD44-expressing immune or stromal populations.
