Product Description
The Cx3Mab-4-CP113 monoclonal antibody is a recombinant, chimeric version of the original Cx3Mab-4 antibody. The variable domain sequences are identical, but the constant region sequences have been switched from Rat IgG1, κ to Mouse IgG2a, κ for use in murine models. Additionally, Cx3Mab-4-CP113 includes the Fc silencing mutation variant LALA-PG, rendering it unable to bind endogenous murine Fcγ receptors or C1q to induce antibody-dependent cell-mediated cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC). The LALA-PG variant has demonstrated significantly reduced effector function, C1q binding and C3 fixation compared to other common silencing mutations such as the LALA and DANG variants while retaining favorable biophysical and manufacturing properties and preserving FcRn-mediated half-life. Species-matched chimeric antibodies demonstrate reduced immunogenicity and formation of anti-drug antibodies (ADAs) compared to xenogenic antibodies in animal models. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.
Mouse CXCR3 (CD183) is a seven‑transmembrane G protein–coupled chemokine receptor expressed mainly on activated T cells, NK cells, and some other immune and stromal populations in mice. It binds the interferon-inducible CXC chemokines CXCL9 (MIG), CXCL10 (IP-10), and CXCL11 (I-TAC), triggering G protein–dependent calcium flux, integrin activation, cytoskeletal rearrangement, and chemotactic migration toward inflamed tissues. CXCR3 is particularly enriched on Th1-polarized and effector/memory T cells, where it supports trafficking to sites of type 1 inflammation and contributes to processes such as allograft rejection and other T cell–mediated immune responses in mouse models.
