Product Description

The YTS 105.18 monoclonal antibody (also referred to as YTS105) reacts with mouse CD8α. CD8 is a transmembrane glycoprotein that functions as a co-receptor for the T-cell receptor (TCR) by binding peptide–MHC class I complexes. It is expressed predominantly on cytotoxic T cells and is also present on subsets of thymocytes, natural killer cells, and dendritic cells. CD8 commonly exists as a CD8αβ heterodimer but can also be expressed as a CD8αα homodimer. Both the CD8α and CD8β chains share significant homology to immunoglobulin variable light chains. The YTS 105.18 antibody recognizes a non-polymorphic epitope on murine CD8α, and it is reported to block CD8 co-receptor interactions with pMHC-I and TL (e.g., MHC-I–dependent CD8 T-cell responses). The non-depleting anti-CD8α YTS105 is shown to synergize with YTS177 and other antibodies in NOD mice by functionally restraining pathogenic CD8+ T cells, supporting combinatorial tolerance strategies across autoimmune and inflammatory models. This recombinant mouse IgG2a LALA-PG variant retains the YTS 105.18 antigen-binding variable regions while incorporating Fc-silencing mutations engineered to reduce Fcγ receptor- and C1q-mediated effector functions. This antibody is useful for Fc-silenced clean CD8 co-receptor functional modulation, CAR T biodistribution/phenotyping, and for CD8-targeted in vivo CAR-T generation via antibody-conjugated tLNP mRNA delivery.
