The YTS 105.18-CP231 monoclonal antibody is a chimeric version of the original YTS 105.15 antibody. The variable domain sequences are identical but the constant region sequences have been switched from rat IgG2a to mouse IgG2a. The YTS 105.18-CP231 has an effector function competent Fc domain allowing for activation of Fcγ receptors (FcγRs) to trigger antibody‑dependent cellular cytotoxicity (ADCC), antibody‑dependent cellular phagocytosis (ADCP), complement‑dependent cytotoxicity (CDC) and opsonization. Species-matched chimeric antibodies result in less immunogenicity and formation of anti-drug antibodies (ADAs) than xenogenic antibodies in animal models. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies.
The YTS 105.18 monoclonal antibody (also referred to as YTS105) reacts with mouse CD8α. CD8 is a transmembrane glycoprotein that functions as a co-receptor for the T-cell receptor (TCR) by binding peptide–MHC class I complexes. It is expressed predominantly on cytotoxic T cells and is also present on subsets of thymocytes, natural killer cells, and dendritic cells. CD8 commonly exists as a CD8αβ heterodimer but can also be expressed as a CD8αα homodimer. Both the CD8α and CD8β chains share significant homology to immunoglobulin variable light chains. The YTS 105.18 antibody recognizes a non-polymorphic epitope on murine CD8α, and it is reported to block CD8 co-receptor interactions with pMHC-I and TL (e.g., MHC-I–dependent CD8 T-cell responses). YTS 105.18 is shown to synergize with YTS 177 and other antibodies in NOD mice by functionally restraining pathogenic CD8+ T cells, supporting combinatorial tolerance strategies across autoimmune and inflammatory models.
