Product Description
CPB513 is a 2+2 symmetric tetravalent bispecific engineered to simultaneously target mouse lymphocyte-activation gene 3 (LAG3) and mouse programmed death-ligand 1 (PD-L1, CD274, B7-H1) proteins. CPB513 contains the murine IgG2a constant region to reduce immunogenicity and the formation of anti-drug antibodies (ADAs) in mouse models and LALA-PG Fc-silencing mutations to abolish antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) while minimizing Fc-driven off-target toxicity. The antibody blocks both LAG-3 (expressed mainly on activated and exhausted T cells) and PD-L1 (expressed on many tumor cells and immune cells), simultaneously inhibiting these immunosuppressive pathways. This bispecific was designed to synergistically target two key immune checkpoint molecules on immune and tumor cells, enhancing activation of tumor-reactive T cells, reversing T cell exhaustion and restoring anti-tumor immune activity and improving anti-tumor response.
