Recombinant human R-spondin 3 (RSPO3) protein is a secreted member of the R-spondin family (RSPO 1–4), containing furin-like and thrombospondin structural domains, that activates the canonical Wnt signaling pathway through the leucine-rich repeat-containing G-protein coupled receptors LGR4–6. In the colon, R-spondin 3 promotes stem cell recovery and epithelial repair by inducing Wnt signaling, and it is an essential component of Wnt3a/R-spondin/noggin (WRN) media used widely to propagate intestinal organoids. As a defined, high-quality recombinant reagent, RSPO3 replaces variable conditioned media (spent media containing Wnt and R-spondin), improving reproducibility and reducing lab-to-lab variability in stem cell and organoid culture.
