Product Description

The TH69-CP247 monoclonal antibody is a recombinant mouse IgG1, κ isotype variant of the anti-human CD7 clone TH69 that reacts with human CD7. The parental TH69 hybridoma was generated at the University of Erlangen-Nürnberg by immunization with the human T-cell acute lymphoblastic leukemia lines CEM and HSB-2, and shows little reactivity outside the T lineage. CD7 is a 40 kDa type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. CD7 is also known as GP40, LEU-9, TP41, and Tp40. It is expressed on the majority of thymocytes, mature T cells, and NK cells, as well as on early lymphoid progenitors, where it is one of the earliest surface markers of T-lineage commitment. CD7 has been reported to interact with SECTM1 (K12) and galectin-1 and plays roles in T and NK cell co-stimulation, adhesion, and activation. CD7 is highly expressed in T-cell acute lymphoblastic leukemia (T-ALL), T-cell lymphomas, and a subset of acute myeloid leukemias (AML), while being largely absent from non-hematopoietic tissues. Because of this expression profile, CD7 is considered an attractive target for antibody-, immunotoxin-, and CAR-T cell-based therapies for T-cell malignancies, where CD7 expression on normal T cells necessitates strategies to avoid CAR-T fratricide. A defining feature of the TH69 clone is that antigen binding triggers rapid internalization of the CD7-antibody complex into the endosomal and lysosomal compartment, a property that has been exploited in scFv immunotoxin, antibody-drug conjugate, and targeted payload delivery formats. TH69 recognizes human CD7 and is not expected to cross-react with mouse CD7, which lacks the proline-rich extracellular repeats present in the human protein. Unlike the original hybridoma-derived clone TH69, clone TH69-CP247 features the TH69 variable region on a mouse IgG1, κ backbone, providing sequence-defined, lot-to-lot consistent material for in vitro and in vivo studies.
