Product Description
This Cibisatamab biosimilar antibody uses the same variable regions as the therapeutic antibody Cibisatamab, making it ideal for research use. Cibisatamab is a humanized IgG1-based T-cell bispecific antibody in a trivalent 2:1 format, containing two CEACAM5-binding arms and one CD3ε-binding arm. The molecule uses kappa/lambda light-chain architecture with domain crossover engineering to promote correct chain pairing, while its Fc region is engineered to reduce Fcγ receptor and complement binding while retaining Fc-mediated half-life extension. By binding CD3 on T cells and CEACAM5 on tumor cells, Cibisatamab redirects T cells to CEACAM5-expressing tumor cells, promoting immune synapse formation, T-cell activation, cytokine release, and targeted cytotoxicity. This T-cell–engaging mechanism enables potent tumor cell killing independent of native antigen presentation and makes Cibisatamab a valuable model for studying T-cell redirection in solid tumor immunotherapy. CEACAM5 (carcinoembryonic antigen) is highly expressed in a range of epithelial malignancies including colorectal, gastric, pancreatic, and non-small cell lung cancers, while expression in normal adult tissues is comparatively restricted, making it an attractive tumor-associated antigen. This Cibisatamab biosimilar is well suited for studying T-cell redirection, CEACAM5-targeted tumor killing, solid tumor immunotherapy, and the mechanistic basis of CD3-engaging bispecific antibody therapeutics.
