Product Description
CPB525 is a 2+2 tetravalent bispecific structured after Ragistomig but designed to simultaneously target mouse programmed death-ligand 1 (PD-L1, CD274, B7-H1) and mouse 4-1BB (CD137). CPB525 contains a murine IgG2a constant region to reduce immunogenicity and the formation of anti-drug antibodies (ADAs) in mouse models and LALA-PG Fc-silencing mutations to abolish antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) while minimizing Fc-driven off-target toxicity and depletion of checkpoint‑expressing effector T cells. This bispecific is engineered to block the PD-L1 checkpoint while delivering conditional 4-1BB co-stimulation in the tumor microenvironment. One arm binds PD-L1 and prevents PD-L1 from engaging PD-1, which removes an inhibitory signal on T cells. The other arm engages 4-1BB (CD137), a costimulatory receptor that boosts T-cell survival, proliferation, cytokine production, and cytotoxic function. This dual-action design aims to combine checkpoint blockade with immune activation.
