Product Description
CPB529 is a 1+1 bivalent bispecific designed to simultaneously target mouse programmed cell death protein 1 (PD-1, CD279) and mouse/human/rat/monkey inducible T cell co-stimulator (ICOS). CPB529 contains the murine IgG2a constant region to reduce immunogenicity and the formation of anti-drug antibodies (ADAs) in mouse models. This bispecific was designed to function as a "molecular switch," effectively shifting the immune balance in the tumor microenvironment (TME) from a suppressed state to an activated, cytotoxic state by combining checkpoint blockade with costimulatory engagement in a single molecule. Blocking PD-1 prevents the delivery of inhibitory signals that otherwise drive T-cell exhaustion and tumor evasion and facilitates T-cell proliferation and cytokine production. Engagement of ICOS provides critical costimulatory signals that drive T-cell proliferation, survival, and the production of effector cytokines like IFN-γ. The anti-PD-1 × anti-ICOS bispecific approach leverages the synergy between removing an inhibitory block (PD-1) and providing a positive stimulus (ICOS), aiming to convert immunologically "cold" or resistant tumors into "hot" responsive environments.
